I’m not sure if you’re asking about WPMU or an option to have the same Shield settings on multiple sites automatically.
If your question was “Can Shield be used in Multisite WordPress (WPMU)?”, then the answer is yes, it can be network activated, but there are no options for subsites.
If your question was “Is there an option to setup the Shield Security plugin on 1 site and have all options replicated to other sites automatically?”, then the answer is yes. There is an option available (“Export/Import” Pro feature). Please see here:

Hello,
Email you received is for “Core Files Scanner”.
You can repair unrecognised WordPress core files automatically – go to Hack Protection module => Core File Scanner (enabled) => enable “Auto Repair” to set the scanner to automatically repair files when the scan next runs.

JelenaYep, custom support like this for 3rd party plugins would only really be addressed for Pro subscribers. We wouldn’t have an eta on it, but best thing to do is subscribe and then contact support directly, and so on.
Also, if it’s going to be working with 3rd party security plugins, the solution would probably be built into the Shield Pro version and not the free, depending on the complexity.

Basically, no, you can’t.
But, in case you have multiple sites and you want to have the same security settings for all of them, you can use automatic “Import/Export” feature. This is how it works:
