This would require some custom coding, and isn’t currently available. Sorry.
It is a great idea though, and something we’ll consider for the future.

I’m just wondering if smush.it could process images locally instead of send o receive to the yahoo servers.
at this time there are no plans to bring this local. We did look into this some time ago and the requirement of the local server with the specialized image processing would be impossible to package into a plugin that would work across a wide range of server configurations.
Recently I check the bulk smush out, which was a really bad experience; and you can’t smush hundrads of images one by one.
Agreed. We are looking at some WP Cron based logic so that you don’t need to attend to the bulk processing yourself. Just upload the images and the scheduled WP Cron will then process the image via the Yahoo! service behind the scenes.

You can please try this beta of Smush.it 1.6.5.4 https://dl.dropboxusercontent.com/u/2616987/WPMUDev/wp-smushit/beta/wp-smushit-1.6.5.4-Beta1.zip
Once installed go to the Settings > Media page and look for the Smush.it section. You will see a new checkbox to turn off the enforce same URL check. Then try either Bulk Smush.it or individual.
This may not work depending on your setup.
The Smush.it plugin should not be effected by the wp ecommerce plugin. When you upload images the Smush.it plugin attempts to use the Yahoo! Smush.it API to have it process the image. This can delay the upload processing.
If you go to Settings > Media on your site you will see there is a section for the Smush.it plugin. There you can turn off the option to have Smush.it process images on upload. If this is turned off you can use the Bulk Smush.it option via the Media menu.
I’m not sure why you would be seeing the 502 HTTP error. It could be that you are not allowed to make external calls from your server or you are running on a local host.
I’m trying to get one of my development sites setup so I can see if there is a way to somehow use the local image. Can you provide some details on your sub-domain setup?
