Yes. It is possible to apply the rule just for one item (1 quantity of the item). You can consider using the Bundle/Set discount type to achieve this.
So the first quantity can get X% discount, and successive quantities will be priced full.

Meanwhile, can you let us know about the “category of users”? Are you referring to user roles ?

Hello and thank you for your quick reply.
For “category of users” we are referring to “user roles”.
We would like to manually create a specific category of users (let’s say “guides”) who have the possibility to buy the first quantity with X% discount and then, successive quantities priced in full.
Ths discount has to be available only one time for every product we sell, then it’s full price.
User “guide” is manually created by us with given roles.
Let us know if it’s possible.

It seems the plugin works.
As I cannot actually buy in demo version, I need to know if, once the first item is bought with applied discount, same user cannot buy the same item again with discount.
How can I set this?

You can use this type of discount to give discount only on the first quantity of the item:

if you also want to prevent the customer from purchasing the same item in the next purchase with a discount, then you can consider using the “Purchase history condition”
https://docs.flycart.org/en/articles/4223893-discount-based-on-previous-purchase-amount-2-0
But, please note that “purchase history” condition only applies at the checkout or when customer is logged (so that the history can be checked).
So using the purchase history rule, you can set it like:
Apply discount only when the customer’s previous order history does not include X Item
