Slug doesn't work

After upgrading to the latest version after I upload the slug won’t change. It automates to the post name. I have tried everything from calling it a Custom Field to adding dashes, to removing dashes and it still comes out as the automatic slug.

Does the permalink setting have to be set to something unique?
