Connections does not do user registrations if that is what you are asking. You can use the builtin user registration in WordPress or a plugin that does user registration. Which one is best really depends on your needs.
If you are asking how to manually add a user, you would do that on the WordPress Users admin page.

So the WP User profile and the individual you create using Connections are two separate entities that are not connected?
I am setup using BuddyPress and while I would like to use this plugin as a directory, I am not yet seeing how it can happen.
Please let me know if I am wrong.

Correct.

You could use the Link extension to link the WP user to their directory entry. This way the user could maintain their entry.
If you setup your WP users first, must have first and last name and email, you could CSV export that data and then import it into the directory using the CSV Import extension.
