No, all the files in the media library must be stored where the site has been configured to store files for the media library. By default, that is in wp-content/uploads. That can be changed according to the instructions found on this page, https://codex.wordpress.org/Editing_wp-config.php.
You can change the location or name of the uploads folder but you cannot change the location of wp-content. Media library Folders would not work correctly if this folder name or its location was changed.

Thanks Alan for you answer. That was exactly what I wanted to know. That’s so a pity it wouldn’t work.
Anyway, may we expect a future version of Media Library where we would be able to perform that? (so we can for instance have http://mybesite/myfolder/)
No, there is not much demand for this type of feature.
