When a new member registers on my site, they’re sent an activation email.
The from email is my-bluehost-username@box-some-numbers-here.bluehost.com
So my question is – How do I change it to, for example, no-reply@mysite.com ?

You can set the email address for your WordPress by logging into your Dashboard as the admin.
Just go to the General Settings page to set the email address.
The other requirement is that this email address must exist in your Bluehost account to use it. So make sure you have it created in your Bluehost cPanel or it will still use username@box#.bluehost.com

Here are a few plugins that allow you to customize the email from name, email etc.

@bh_wp_guru For some reason it didn’t work.

And thanks @rajeshsoni ! WP Mail From II seems like the solution I was looking for!
