Activate the lightbox?

How do I activate the lightbox on a gallery?

I’ve got ‘Add lightbox to WordPress image galleries by default.’ set to enable but nothings happening.

I’ve tried manually adding rel=”lightbox” but again no impact:
[gallery ids="800" rel="lightbox"]

What am I doing wrong?
