BP 1.6 has moved some of the defines to filters. Are you still on BP 1.5*?

it might be because the current version is for BP 1.6. Try reverting back to old version http://wordpress.org/extend/plugins/buddypress-stalker/developers/

The menu needs to be created as well. Go to the menus admin and create a menu and set its location to the mobile menu.
