The link generated is correct, so it doesn’t seem tough to provide specific meta data for a Twitter card. I will proceed to code this myself unless somebody found a reason why it won’t work…
It just surprises me this wasn’t built in. Which makes me think maybe there are issues…

Instead of hard-coding the gallery, use the “Attach Nextgen Gallery to Post” option when posting something in WordPress (one of the icons in the visual area). From here you can specify all kinds of options, it’s really cool and works great.
Yes, I am using random and one specific gallery.
