Does this plugin support Nginx reverse proxy mode? Or, do we have to manually convert and add the rewrite rules for static files in Nginx configuration file?

You can of course use nginx as a reverse proxy, but W3TC doesn’t do any configuration of it. Using nginx as the web server does take advantage of configuration files W3TC creates.
