Getting a Internal Server error 500 after setup

I just installed WP and a new Theme. Everything so far. Then I installed the iThemes Scecurity Plugin and made some adjustments within the Dashboard (no code changed). Apparently the created (or automatically changed) htaccess file created an error 500, and no page could be viewed anymore.
Putting the code of the htaccess file into a code checker brought up no mistakes in the syntax. BUT when I delete the htaccess file the site is working again.
Has anyone an idea what could be wrong within the htaccess file??
