Yes, or you will see an issue at some point. You only want to be running one minify option on a site at a given time.

You shouldn’t have any downtime switching to our nameservers.
I assume you’re using apps for mail. This will also work fine as long as you have the correct mail and mx records in your zone file for the domain.

the advice seems to pertain to mydomain.com – what happens if my site url is http://www.mydomain.com – what kind of pagerule do I create?

Yes, if you want it to wildcard. Say you’re doing one for the wp-admin section, for example, it would cover something like:
yoursite.com/wp-admin/
yoursite.com/wp-admin/adifferenturl
