Could I use a multisite network with domain mapping a subdomain and identify different languages in a directory structure like below for example with your plugin?

sub.domain.com (English Main Language)
sub.domain.com/es (Spanish)
sub.domain.com/fr (French)

Yes, that should work. Multilingual Press gets its URLs from WordPress per API functions, any changes made by the mapping plugin are respected. But please test it on a local test installation first, just to be sure.
