Conditional tags in post

I need to add the conditional tags is_single() and is_home() into certain posts to toggle the auto-play of a video on single and home pages. This isn’t working for me, and a quick google search didn’t turn up anything on whether these tags can be used in the context of a post. If not, any ideas on how to do this otherwise?
