The first option (Auto-update “Current URI”) can be used to make my plugin automatically update the actual permalink, when the default permalink is changed.
The default permalink is displayed next to “Default URI” field. In short, the default permalink is based on the permastructure format that can be changed in “Tools -> Permalink Manager -> Permastructures” section.
Please keep in mind that if “auto-update” mode is turned on, your manual changes to the custom permalinks can be overwritten.

As regards the last option (“Automatic redirect”) it is useful if you would like to provide a fallback for old/original permalinks (used before my plugin was installed):
