I do have exact the same problem at my WP installation. At my site all galleries or albums are affected.
Problem appeared yesterday in the evening without any changes at wordpress or the plugin configuration.
WP: 2.9.1
Plugin Ver.:1.4.3

This would suggest an external JavaScript which is called was changed.
Here’s what I see as possibles:
ajax.googleapis.com/ajax/libs/swfobject/2.2/swfobject.js
lite.piclens.com/current/piclens_optimized.js
Any thoughts?

I have been having exactly the same problem – the plugin was working fine, then suddenly the lightbox style gallery stopped functioning and the thumbnails now just link to plain pictures like yours do Carsten. Same with me – no changes to WP or the plugin or any other changes behind the scenes.
Hope someone can help with this – I was planning on using this plugin on a couple of sites I’m currently working on 🙁

I fixed the problem at my site, although I don’t understand the way I fixed it. I use at my site also the Tweet Blender Plugin. And I noticed that the Tweet Blender did not update the tweets since the problem with NextGen Gallery appeared. So I played with Tweet Blender settings and I cleared the cache of Tweet Blender. After reloading the page tweets was updated and, oh wonder, NextGen Gallery worked again.
I don’t have a glue if this helps any of you, but I wanted to report…

sounds similar to a problem that developed for me towards the end of last week….
still cannot get it to work again…
with 4000 pages and over 8000 images i dont want to have to re-edit the pages
