The old method of doing this was to edit wp-includes/kses.php to restrict the markup allowed in comments but you might want to look for a plugin that can help you do this more easily. Another option would be to hold comments for moderation if they contain only 1 link via Admin/Settings/Discussion. But how practical that might be, I couldn’t say.

Is there anyway to script/code a requirement that comments with links cannot be allowed?
