How do I create a single.php file?

I’d start by making a copy of index.php, then remove references to comments_popup_link and add <?php comments_template();?> just before the <?php endwhile; ?> of the main Loop.

That should get you started with a basic single.php file – after which it’s just a case of tweaking it to suit your tastes.
