    WordPress is a tool I frequently involve for my customers' web needs. I'm starting to think about a way to simplify usual maintainance operations, in order to centralize installations, updates and backup activities.

    I'd like to setup a SINGLE WordPress installation, able to serve many different customers, each one with his own URL, his own theme, hiw own contents. This system should be able to enable or disable plugins for each customer, based on each one needs.

    This way I could greatly enhance all the operations now fragmented into many many different websites.

    Is there a way to setup such a system with WordPress?

    I have a personal web server + web space I usually resell to customers in order to host their sites.

    WordPress mu with Donncha's WordPress MU Domain Mapping plugin should be able to do what you want. The functionality of the WPmu project will be merged into WordPress.org in v3.0 which is expected to be released in April.
