That involves setting up your home.php (Codex ref. Template Hierarchy), setting up the divs with either dynamic sidebars or directly coding functions onto template or mix of both, and adding the necessary CSS in your style.css.
To get some ideas, check out the layout’s source code and css. But if that’s something that’s beyond your scope, Google “Free WordPress templates” and find something that approximates the look you want and change background colors, font sizes etc. to your liking in style.css.
Finally, you can always hire a web designer who’s versed in WordPress.
