Select Specific time to have busy or block on a specific day.

You can set Exceptional working or non working days in Appointments -> Settings -> Exceptions.

However, you can’t set exceptions for specific times on that day but only for days.

I’ve also found an interesting workaround that you could use given by one of my colleagues on WPMU DEV forum here: https://premium.wpmudev.org/forums/topic/appointment-setting-times-for-exception-days#post-549740.
