Camer/microphone access would be related to “Feature Policy” security header which Defender doesn’t currently set/use.
If you are absolutely sure that this happens due to Defender, then you can try disabling “Permissions-Policy” (temporarily for testing) on “Defender -> Tools -> Security Headers” or test them by disabling them one by one to find out which one is causing that (but if any of them it would rather be a “side-effect” due to some other aspects of site configuration rather than actual cause).
However, I think this would be set by something else (either other code or your host). Are you able to share a link to the page in question so we could take a look from outside?

Yeah it’s definitely just defender making it happen. I found out that Permissions-Policy is causing it. How can find out what I should insert into specific origins to use that security feature but to have my video calls working?

You can set it to Specific Origins in case you would like to limit it to the domain, but the rules really depend on your security requirements.
