If you move the site you would have to set up domain forwarding so the subdomain name still points to the site. Unless you use a different domain also-

Right now I am working on the site
I have put the wordpress in suddomain.mysite.com in another hosting
and in my main site I just have htaccess and index.php copied
but I get 500 error

You can use the subdomain with another hosting provider so long as you have the DNS records set up correctly to point your subdomain.site.com to the IP address of the new hosting provider. You can still have site.com work on the current hosting provider using the dns records for that host.
Any links from site.com that take you to subdomain.site.com will work once that setup is completed.
A 500 error could be a number of things, more common these days is an invalid version of PHP running that your site is not compatible with. Verify the PHP version you are running with your current host and compare that with the new one to see if it is lower or higher than your current provider.
