I am not sure what the issue is. I have not been able to get reproducible behavior. What I can say is that prior to the update – I could send an email to my site and it would ALWAYS show up as a post. After the update it is hit and miss sometimes it works – sometimes it doesn’t. When I run the Test Config what I get is posted below. It shows that “# of waiting messages: 1”. I can run Process Email and it immediately works.

Postie is totally dependent on the WordPress cron working. So it emails aren’t begin picked up that typically means that cron isn’t working.
I notice you have alternative cron turned on which may or may not be related.
The other option is to use an external cron. See http://postieplugin.com/forcing-an-email-check/
You might also look at your site health (Tools->Site Health)
