I need a little help how to cache directories and certain file extensions.

If I wanted to cache all my uploads, would I cache:
wp-content/uploads
wp-content/uploads/
wp-content/uploads*
wp-content/uploads(.*)

This is not possible. Let me ask why you want it 🙂 Generally there are static files in uploads folder.
