I am testing this on a clone of my site, is there some sort of drip feed way I can kick this off, you page says it is automatic?

Is is automatic for new images only?

It could just do them between two dates or something

I have about 3600 images already uploaded which means I have 5 times that because WordPress makes so many sizes.

I went to the media library and tried to select 500 and it said

    “Request-URI Too Large

The requested URL’s length exceeds the capacity limit for this server. “

So how many can I do at a time?

This will take forever if I have to do it all manually

Yes, it is automatic for new images only, which is why there is a Bulk Optimize option in the Media Library to do all of them at once.
If I remember correctly, there is a limit in php or apache to how long the request url can be, but if you use the normal Bulk Optimize function, it allows you to do your entire library (my test library has 2300, and I run it pretty frequently).

What do you mean by the “normal” Bulk Optimize option

I changed the settings clicked save, not seen any other things, so I went to the media library where I had set the screen option to 500, selected all and tried to bulk optimize, that is when I hit the error above.

Is there some other hidden “normal” option I am missing?

In the WP menu under Media, there is a Bulk Optimize option. Should be directly under ‘Add New’
