If you have askimet and either bad behaivor or spam karma installed then your pretty safe to just accepting comments.
If you look in your dashboard under Options then under Discussion, all your comment options are there. You can disable an administrator must approve a comment before it appears, you might want to check Comment author must fill out name and e-mail.
Also you might want to have a look at the comment moderation options. Especially the holding a comment with more then X number of links. Usually spammy comments will have more the 3 links.

Is there a way to auto accept comments without having to manually doing it yourself? I’d still like to keep the spam out.
