I’m using Subscribe2 6.5 on WordPress 3.2.1 and have been doing since WordPress 3.2.1 was released in July.
It works on both of my sites – one in per-post mode and one in digest mode.
Try disabling other plugins, revert to the default theme and make sure you are using the most recent version of Subscribe2 (version 6.5).
6.5 was working up until wp 3.2.1 update. none of the usual troubleshooting methods have been successful. I noticed that 5 of 10 users who have rated the 6.5 plugin version are also saying it is “broken”. I am reverting to 6.4 to see if the issue is indeed with the update.

If you can find a solution please let me know what it is so I can see if anything needs to be amended in the Subscribe2 code.
I find it strange that you say Subscribe2 6.5 was working just fine until you updated WordPress to 3.2.1 and yet now the plugin is not working as expected it’s an issue with the plugin – even though that has not been changed.
