Using WP 3.0.3, and Postie 1.4.1, and when I try to run /wp-content/plugins/postie/get_mail.php either by cron job, by hand, or even manually, I get a 500 error. Nothing shows up in the server logs either.

Try to disable "Show friendly HTTP error message" from IE > Tools > Options > Advanced. Then you'll be able to see the error. :)
I'm not using IE. In fact, same error on my Ubuntu desktop.
The IE thing was just to figure out the error message...
Try to access the page by adding this parameter: content/plugins/postie/get_mail.php?Submit=Run+Postie
That gives me a 404, Page Not Found.
I get a 500 internal server error when I try to activate the plugin. Any ideas why that might be?
