- make sure your plugin version is compatible with WP3.2.1
- If it is, try deactivating ALL plugins temporarily to narrow down the problem. If the problem goes away, activate them individually (one-by-one) to find the offending plugin.
I only have NextGen active.
I'm currently working on a similar problem and have found that if you upload images through FTP they might belong (as far as the server is concerned) to a different "user" than the "web user" that you use through the browser. I think my biggest problem is that the server runs on Windows/IIS 7 :)
Did you upload through FTP or use NextGen's uploader?

I did not use FTP.
I poked around and it seems that NextGen does not like very large images. The ones I uploaded were on average 4000 x 3500 and once I resized them to 1920 x 1240 thumbnails were generated. So in future I'll be more careful with the size of images I upload.
I was having the same issue. I tracked it down to a conflict with wp_supercache. It may or may not be that for you, since I haven't found anyone else with the same problem, but I just disable that plugin when I'm uploading and re-enable it when I'm done.
Either way, it is most likely a plugin conflict.
I Just uploaded 14 images that I resized to 800x600 pixels.
I cannot get any thumbnails to generate?? When I click the X I can see the photo but there are just X 's on my site where the thumbnails should be??

How do I get the thumbnails to generate?? If I click on EDIT THUMBNAILS all I see are lines of code with fatal error messages and the 2 boxes below just have the X in a white box??
