Will this work with a regular wordpress3.0 (NOT mu) installation with NO mu-plugins folder and subfolders multisite activated?
That would be great
If you;re running multisite it’s the same thing as what was mu.
Create the mu-plugins folder. WP will recognize it.

1. it did not appear in the plugins list (nice)
2. its options were available only for the super-admin (nicer)

Generally speaking, yes.

Look at the plugins sub-menu items. π There will be one called “must-use”. Only Super Admins see this.

No. The plugin itself has to be coded that way. It’s not a function of the folder itself.

works on ‘blog.example.tld’ site, and ‘example.tld/blog’ network setups!
