I'm wanting to change the layout on selected pages on my blog. For example, the default layout for my site is a 2 column look, but I want a single column look for one particular page. Can this be done?

Do you mean Page as in WordPress Page, or do you mean it more generically?

A WordPress Page can be assigned its own template:

If you need to set a post to an unique template, see this plugin:

Beyond these options, with a little knowledge of PHP and WordPress' conditional tags, you could modify any theme's templates to alter its layout based on the query type (post, Page, or whatever).
