Aside from adding each (new) subdomain manually to the /etc/hosts file, are there any other workarounds to sidestep the wildcard DNS requirement for running WP 3 in multisite mode on localhost (like under apache / XAMPP) ? Thanks!

Nope, just adding them manually in your hosts file.
