I've got MultiSite successfully installed in MAMP, but I'm unable to use its sites from other computers on my local network. Because the MultiSite name is 'localhost', the remote comp tries to load assets from localhost (itself).
Is there a basic adjustment I'm missing that would allow this to work properly? I'm assuming setting up proper DNS with a domain and hostnames would solve it?

It's not really a WordPress (or Multisite) issue :) The problem is your site isn't 'local' to other computers, as you've guessed. You might be able to use your IP address instead of localhost, to do that.
