How do I make this work because I added <a href="”/images/main/123.jpg”" title="”my">test</a> and it doesn't do anything.

Add the attribute: rel="lightbox" to your link tag.
Make sure the lightbox plugin is activated in the admin panel.
This may be of some help if that fails.
