Wordpress is installed in a sub-directory. So I access it at "http://mydomain.com/wordpress"
Can I set it up so that when I go to "http://mydomain.com" that it loads wp-login.php
and after it takes the user and pw go immediately to the home page of the blog.
If this is possible, what files and changes do I need to make?

if you have control panel 10 , then you can set up a permanant divert, so you divert "www.yourdomain.com/wordpress/" to "www.yourdomain.com" so when people click the yourdomain.com link there divert directly to your /wordress/index.php file, ie: your blog ;) this is the most easiest and best way to do it, however there is another way which involves moving a few files, but this will interfer with certain themes AFAIK
