It’s probably a conflict with an older plugin of mine.
1. Deactivate all plugins.
2. Activate P2P.
3. Activate all the other plugins, one by one.

Hmmm, with plugins off, I got a different error.

Are you sure you deactivated ALL of them? No network-activate plugins? No files inside mu-plugins?
Also, maybe some files were not downloaded when you upgraded. Make sure you have an scb
directory inside the posts-to-posts directory and that it contains all the files listed here:

Hi Scribu, I was in the same problem, and I detected that it has incompatibility issues with your WP-PageNavi plugin, uhmmm… maybe because it loads your “scb” library twice when both plugins are installed?

Yeah, the problem was that it was loading the older version of the library from PageNavi. Should be fixed in the development version (1.1.1-alpha).

Ah, I came back to mention WP-PageNavi and there’s a new release! I think I had an additional problem today in that W3TC object cache is enabled in the admin here due to some dashboard feeds that need to be cached. When I cleared cache after disabling WP-PageNavi tonight, I was able to install v1.1. Anyway on to v1.1.1.
By the way congrats on reaching the 1.0 milestone, even though that is now old news, it is the reason we are evaluating this very useful plugin.
Tomorrow, I want to get WP-PageNavi working with Post2Post on a master-detail page, I’ll share the results.
