Here is little REQ for future development of WP: possibillity to run multiple blogs (plus) with multiple (different) URLs - all under one instance of WP.
This would be important improvement. Such feature/s has NucleusCMS and it is much liked by thier users.
Why not give it to WP community?

Oh, there have been attempts. What you're talking about has been tried here, and also here. I've been down that road and haven't had much success, though some have.
