I started managing a podcast as a subset of my site http://www.SciFiDimensions.com. WordPress is installed in a folder called "podcast" (It's at http://www.SciFiDimensions.com/podcast). Now I'd like to use WordPress as my main content manager, and I'd like to have that "podcast" folder be named "main".
If I just go in and change that folder name, will it screw up the existing blog/podcast entries? What's the best way to make the transition?

Yes! Big time.
1. admin > Settings > General
2. Change the two URL values to the folder name you want
3. Save and disregard the error message!
4. Don't try to see your blog in a browser!
5. Via FTP move rename the podcast folder to what you changed it in settings
6. Should work

Thanks I will give it a shot.
Now...I assume this means that anyone who's hardlinked to a specific article from the outside will lose that link, yes?

Yes, but you can use a 301 permanent redirect for that in a htaccess file.

Google, there are many sites explaining it - it is NOT a WP issue.

Another step:
2a. Admin > Settings > Miscellaneous
Update the path to the uploads folder
