Update:
I removed all of the former WP files and copied over all of the 1.0.1 files. Now when I use upgrade.php, I get this too:
There doesn't seem to be a wp-config.php file. Double check that you updated wp-config.sample.php with the proper database connection information and renamed it to wp-config.php.

I'm getting the same errors as debwire after uploading the new files and running upgrade.php.
Everything still seems to be working ok though. :-/
