If I upgrade to Wordpress 2.1, will my current theme still work? It's called Vistered Little 1.6 and a theme on my other blog called Digg 3 Columns 1.0. Will those work with the new version of Wordpress?
I also have "Add Meta Tags" plugin http://www.g-loaded.eu/2006/01/05/add-meta-tags-wordpress-plugin/
installed.
Will those be compatible with the new version or will I have to wait for new versions of them to be made?

Neither are all inclusive, but they may help.

The theme I'm using is not listed, but the plugin is. Anyone else have any idea whether or not the theme is compatible?
If I do go ahead and upgrade, can I downgrade back if the theme doesn't work? I'm really attached to it.
Downgrade? No.
But why not just back it all up properly BEFORE the upgrade. if you have problems, you just restore the backup.
Have a look at WordPress_Backups for more on how to backup files and database.
